KNU Research Team on Endocrinology and Metabolism Publishes Paper in Top Research Journal

Kyungpook National University’s Research Team on Endocrinology and Metabolism (Corresponding Authors: In-Kyu Lee, Jae-Han Jeon; Co-first Authors: Ji-Min Lee, Min-Ji Kim) successfully published a paper in one of the most highly regarded journals on bone and mineral research, The Journal of Bone and Mineral Research (JBMR, IF 5.854).* The paper, “PDK2 Deficiency Prevents Ovariectomy-Induced Bone Loss in Mice by Regulating the RANKL-NFATc1 Pathway During Osteoclastogenesis,” was published on the front page of the journal’s March 2021 Edition.

 

The paper states that estrogen deficiency increases the osteoclastic resorption of bone, leading to osteoporosis (bone disease resulting from a decrease in bone density). It also claims that because during osteoporosis, there is an increase in mitochondrial enzyme pyruvate dehydrogenase kinase (PDK2), an inhibition of PDK2 would prevent osteoporosis by reducing aberrant osteoclast activation. Indeed, experiments showed that in estrogen-deficient mice, an inhibition of PDK2 prevented the bone-resorbing activity of osteoclasts through a reduction in NFATc1 expression and thereby prevented ovariectomized(OVX)-induced osteoporosis in mice.

 

The research team states, “This paper presents evidence that PDK2 inhibitors may be a key factor in the treatment of osteoporosis induced by estrogen deficiency.”
